Towards a better understanding of the pathogenesis of knee osteoarthritis(更好地理解膝关节骨关节炎的发病机制)

报告内容简介:

Knee osteoarthritis (KOA) is a prevalent form of chronic joint disease associated with functional restrictions and pain in the old people. And it is estimated to affect more than 240 million people worldwide. At present, the main treatment mode for KOA is focused on alleviating symptoms, showing small to moderate effects. Therefore, prevention of KOA is critical to reduce the burden of disease.

KOA is the most common degenerative disease and one of the main causes of severe disability in daily life. It has a multifactorial etiology characterized by pathological changes in the knee joint structure, including cartilage erosion, synovial inflammation, subchondral sclerosis, and bone hyperplasia. The main characteristics of pathological changes of KOA include: proinflammatory cytokines, matrix metalloproteinases and prostaglandins destroy cartilage and promote catabolic environment; Subchondral bone remodeling and absorption; Hypertrophic differentiation of chondrocytes; Synovial tissue neovascularization; Focal calcification of articular cartilage and so on. Previous studies have suggested that the role of muscle properties, blood vessels (vascular), and meniscus pathologies in KOA. However, the relationship between muscle characteristics and vascular pathology with knee joint structural progression and functional outcomes has not been investigated in the elderly population in the community. Meniscal pathology (tears and extrusion) plays an important role in the pathogenesis of KOA. Degenerative meniscal changes are common, but causes for this remain unclear. Given the evidence for an increasing role of vascular pathology in knee OA, the role of statins warrants investigation given the recent data supporting a beneficial role for statins on other knee structures such as bone marrow lesions.

The main purpose of my thesis is to better understand the role of muscle, vascular factors and meniscal pathology in the pathogenesis of knee OA.

曾梦杰,本科毕业于华北理工大学的针灸推拿学专业,硕士毕业于Macau University of Science and Technology的中医学专业,博士就读于澳大利亚Monash UniversityQS42)的临床医学专业。以第一作者在GerontologyThe Bone and Joint JournalFrontiers in Neuroendocrinology等国际知名期刊发表学术论文多篇。
